Resumo O estudo objetivou avaliar os aspectos anatômicos e o comportamento fisiológico de sementes de Peltophorum dubium (Spreng.) Taub. durante o armazenamento, submetidas à ação de tratamentos para superação da dormência. Os frutos maduros foram beneficiados e as sementes armazenadas à temperatura de 5ºC por um período de 360 dias e, posteriormente, submetidas a tratamentos para superação da dormência: (T1 - sem escarificação, T2 - escarificação mecânica e T3 - escarificação química com ácido sulfúrico por 10 minutos). Em intervalos pré-estabelecidos, as sementes foram avaliadas por meio da análise anatômica do tegumento e pelo teste de germinação. As variáveis analisadas foram anatomia do tegumento, porcentagem de germinação, índice de velocidade de germinação e correlação de Pearson entre a espessura do tegumento e as variáveis fisiológicas. Para as análises anatômicas, as sementes foram fixadas, inclusas e seccionadas em micrótomo. A espessura do tegumento das sementes foi mensurada por meio de imagens das secções transversais. O teste de germinação foi conduzido com quatro repetições de 25 sementes, realizado em papel germitest à temperatura constante de 25ºC com fotoperíodo de 12 horas. O delineamento experimental foi inteiramente casualizado em esquema fatorial 5x3 (lotes armazenamento e tratamentos para superação da dormência) e as médias comparadas pelo teste de Skott-Knott a 5% de probabilidade. As análises anatômicas demonstraram perda de resistência física das estruturas do tegumento ao longo do armazenamento. A porcentagem de germinação durante o armazenamento não diferiu significativamente entre os tratamentos, entretanto o índice de velocidade de germinação aumentou e forma significativa quando associado a tratamentos de superação da dormência (T2. 7,08 - sementes não armazenadas e 9,29-armazenadas 360 dias; T3. 6,01-sementes não armazenadas e 9,42-armazenadas 360 dias). Foram obtidas correlações significativas entre a espessura do tegumento e as variáveis fisiológicas analisadas. O armazenamento, por até 360 dias, promoveu modificações anatômicas tegumentares, manteve o alto percentual germinativo das sementes de Peltophorum dubium e, quando associado aos tratamentos de superação da dormência, facilitou a ação destes a partir de 180 dias de armazenagem, aumentando o índice de velocidade de germinação.

Abstract The objective of this study was to evaluate the anatomical aspects and the physiological behavior of seeds of Peltophorum dubium (Spreng.) Taub. during storage and while undergoing treatments to overcome dormancy. Mature fruits were collected and seeds stored at 5ºC for a period of 360 days and later submitted to treatments to overcome dormancy: T1 - without scarification, T2 - mechanical scarification and T3 - chemical scarification with sulfuric acid for 10 minutes. At pre-established intervals, the seeds were evaluated through an anatomical analysis of the tegument and a germination test. The variables analyzed were tegument anatomy, germination percentage, germination speed index and Pearson correlation between tegument thickness and physiological variables. For the anatomical analyses, the seeds were fixed, embedded, and sectioned on a microtome. The thickness of the tegument was measured from the images of the cross sections. The germination test was conducted with four replicates of 25 seeds, carried out on germitest paper at 25ºC with photoperiod of 12 hours. The experimental design was completely randomized in a 5×3 factorial scheme (storage and overcoming dormancy treatment groups), and the means were compared by the Skott-Knott test at 5% probability. The anatomical analysis showed loss of physical resistance of the tegumentary structures during storage. The percentage of germination for the treatments during storage did not differ significantly, however, the rate of germination showed a significant increase when associated with treatments for overcoming dormancy (T2: 7.08 - seeds not stored and 9.29- stored 360 days; T3: 6.01-seeds not stored and 9.42-stored 360 days). Significant correlations were obtained between the thickness of the tegument and the physiological variables analyzed. Storage for up to 360 days promoted tegumentary anatomical modifications, maintained a high germination percentage of the seeds of Peltophorum dubium, and, when associated with treatments for overcoming dormancy, facilitated their action from 180 days of storage, increasing the rate of germination.

RESUMO Mimosa flocculosa Burkart é uma espécie nativa, utilizada para recuperação de áreas degradadas. Porém, a produção de mudas em larga escala é dificultada pela dormência tegumentar presente na semente. Esse trabalho avaliou a germinação e a anatomia do tegumento de sementes submetidas à tratamentos de superação da dormência. Sementes de M. flocculosa foram submetidas aos tratamentos: controle, água aquecida a 50, 80 e 100 ºC e H2SO4 durante 1, 3 e 5 minutos. A germinação contou com quatro repetições de 25 sementes em delineamento inteiramente casualizado. As variáveis analisadas foram: germinação, tempo médio e índice de velocidade de germinação. Foram realizadas ainda análises anatômicas do tegumento das sementes. Os tratamentos usando água a 80 ºC, e ácido sulfúrico por 3 e 5 minutos promoveram mudanças anatômicas benéficas no tegumento da semente, refletindo no aumento da porcentagem e velocidade de germinação. Recomendamos a água aquecida a 80 ºC pela eficiência e maior segurança na manipulação.

ABSTRACT Mimosa flocculosa Burkart is a native species used for the recovery of degraded areas. Large scale seedling production in nurseries is hindered due to seed physical dormancy. This study evaluated the germination and seed coat anatomy of M. flocculosa seeds subjected to dormancy overcome treatments. Seeds of M. flocculosa were subjected to: control with distilled water, heated water at 50, 80 and 100 °C, and H2SO4 for 1, 3 and 5 minutes. Germination tests were conducted with four replicates of 25 seeds in an entirely casualized delineation, analyzing germination percentage, mean germination time and germination speed index. Moreover, anatomical analysis of the seed coat was conducted. The treatments with heated water at 80 ºC, and sulfuric acid for 3 and 5 minutes promoted benefic anatomical alterations in the seed integument, reflecting in higher and faster in germination rates. Heated water at 80 ºC is recommended because of its efficiency and safety.

A romäzeira (Punica granatum L.) é fruteira de porte arbustivo, utilizada geralmente como planta ornamental e explorada quanto a suas características medicinais. Seus frutos apresentam cavidades internas onde estäo alojadas numerosas sementes e sem a presença de endosperma. Estas sementes apresentam certa dificuldade para germinar, sendo que se conseguiu atingir porcentagens satisfatórias de germinação com a utilização de estratificação, que é processo necessário para a quebra da dormência fisiológica, já que altera o balanço hormonal, sobretudo os níveis de giberelina. O trabalho teve por objetivo avaliar a aplicação de concentrações de GA3 na germinação de sementes de Punica granatum em ambiente controlado com temperatura de 25ºC e fotoperíodo de 12 horas de luz, sobre papel germitest. Adotou-se o delineamento inteiramente casualisado, com cinco tratamentos [concentrações de GA3 (0; 100; 200; 300 e 400 mgL-1)], quatro repetições de 25 sementes cada. Determinou-se durante a embebição a curva de aquisição de água das mesmas. As variáveis avaliadas foram porcentagem de germinação, tempo médio de germinação, índice de velocidade de germinação, índice de sincronização e frequência de germinação, comprimento médio total (parte aérea + parte radicular), massa da matéria fresca e seca das plântulas. A única variável que mostrou efeito das concentrações de GA3 foi o índice de velocidade de emergência. O tempo de embebição necessário para atingir máximo teor de água nas sementes de romäzeira (P.granatum) em água deionizada foi de quatro horas, sendo a embebição neste líquido o método eficaz para o processo germinativo, sem a necessidade de uso de giberelina.

The pomegranate (Punica granatum L.) fruit tree is shrubby, often used as an ornamental plant and operated as its medicinal characteristics. Its fruits have internal cavities that host numerous seeds and without the presence of endosperm. These seeds are germinating some difficulty, and it was possible to achieve satisfactory percentages of germination using laminating process that is required for breaking of physiological dormancy, as alters hormone balance, particularly gibberellin levels. The study aimed to evaluate the application of GA3 concentrations on germination of seeds of Punica granatum in controlled temperature of 25ºC and a photoperiod of 12 hours of light on paper germitest environment. We adopted the completely randomized design, with five treatments [GA3 concentrations (0, 100, 200, 300 and 400 mg L-1)], four replicates of 25 seeds each. It was determined during the acquisition curve imbibition of water from the same. The variables evaluated were germination percentage, mean germination time, germination velocity index, index of synchronization and frequency of germination, total length (shoot + root), fresh weight and dry weight of seedlings. The only variable that showed the effect of concentrations of GA3 was the emergence rate index. The soaking time required to achieve maximum water content in the seeds of pomegranate tree (P. granatum) in deionized water was four hours, this soaking liquid effective method for the germination process, without the need to Gibberellin.

O objetivo deste trabalho foi avaliar o efeito de diferentes teores de água atingidos pelas sementes de Annona emarginata (Schltdl.) H. Rainer durante a imersão em soluções de GA3, na variação dos teores de açúcares solúveis e na germinação. Sementes com teor de água inicial de 10% foram colocadas para embeber em soluções com as concentrações de 0; 250; 500; 750 e 1.000 mg L-1 de GA3, e ao atingirem os teores de 15%, 20%, 25%, 30% e 35% de água, procedeu-se à quantificação dos teores de açúcares solúveis e ao teste de germinação. As sementes imersas até atingirem 15% de água com a adição de GA3 e imersas até a aquisição de 20% de água sem a adição de GA3 apresentaram maiores teores de açúcares solúveis e porcentagens de germinação, os quais foram reduzidos à medida que as sementes atingiram 30% e 35% de água, independentemente do uso do regulador vegetal. Conclui-se que os diferentes teores de água alcançados pelas sementes nos tratamentos de imersão afetam os teores de açúcares solúveis e a germinação de sementes de Annona emarginata. Assim, em tratamentos com Annona emarginata, as sementes devem permancer imersas na água sem a adição de GA3 até a aquisição de 20% de água, uma vez que elevados teores de água (35%) reduzem os teores de açúcares solúveis totais e a porcentagem de germinação das sementes.

The objective of this study was to evaluate the effect of different water contents achieved by Annona emarginata (Schltdl.) H. Rainer seeds during immersion in GA3 solutions, in variation of soluble sugars levels and germination. Seeds with 10% of initial water content were submitted to imbibition in GA3 solutions with concentrations of 0; 250; 500; 750 and 1000 mg L-1 and when they reached the water content of 15%, 20%, 25%, 30% and 35%, the quantification of soluble sugars levels and germination test were performed. Seeds immersed up to they reach 15% of water with GA3 and immersed up to the water acquisition of 20% without GA3, presented higher soluble sugars levels and germination percentage, which were decreased when the seeds reached 30% and 35% of water, independently of the presence of the plant growth regulator. It was conclude that different water contents reached by the seeds in immersion treatments with GA3 affect the soluble sugars levels and germination percentage of Annona emarginata seeds. Thus, in treatments with Annona emarginata, the seeds must remain immersed in water without GA3 up to they reach 20% of water, as higher water contents (35%) reduce the soluble sugars levels and the seed germination percentage.

Annona emarginata (Schldtl.) H. Rainer ("araticum-de-terra-fria") is used as a rootstock for several species of Annonaceae. It is suggested that these seeds should be sown immediately after extraction and, therefore, they could be intolerant to desiccation. There are several mechanisms involved with desiccation tolerance. Soluble sugars, for example, can accumulate and act as osmoprotectants for the membrane system during desiccation. The aim of this study is to assess desiccation tolerance in A. emarginata seeds. In addition, we examined the soluble sugars involved in desiccation tolerance. Finally, we determined the effect of gibberellic acid (GA4+7) and N-(phenylmethyl)-aminopurine in promoting the germination of seeds with different water contents. The experiment consisted of a randomized 4x5 factorial design (desiccation levels x concentration of growth regulators). After drying, seeds containing 31 (control), 19, 12 and 5% water were incubated in different concentrations of GA4+7 N-(phenylmethyl)-aminopurine (0, 250, 500, 750 and 1000 mg L-1) for 60 hours. The experiment was conducted in a germination chamber with alternating temperature and photoperiod of 20ºC for 18 hours of darkness and 30ºC for 6 hours of light. We analyzed electrical conductivity, germination rate, mean germination time, germination speed, frequency and uniformity of germination, percentage of dormant seeds and soluble sugar profile in intact seeds through high-performance liquid chromatography (HPLC). The data were subjected to analysis of variance, and the means were compared using Tukey's test at a threshold of p<0.05. The results showed that seeds of A. emarginata appears to be desiccation tolerant and, also, that sucrose increases when seed water content is reduced to values as low as 12%, exogenous GA4+7+N-(phenylmethyl)-aminopurine improves its germinability.
